Comparison of Nanoparticle-Targeted Delivery vs. Bioconjugate Approaches

Hossen et al. (2013) conducted a comparative study in a diet-induced obesity mouse model examining prohibitin-targeted nanoparticles encapsulating the proapoptotic peptide KLA versus the bioconjugate form (adipotide). Measured endpoints included serum leptin and adiponectin concentrations and ectopic lipid accumulation in liver and muscle tissue. The nanoparticle formulation was reported to show greater changes in these measured endpoints than the bioconjugate. The authors attributed this difference to multivalent targeting and enhanced cytoplasmic delivery via endosomal escape in adipose endothelial cells. This study provides context for adipotide as a reference bioconjugate in targeted delivery research; however, findings from a single nonclinical model should not be generalized [Hossen et al., 2013].
